I just wanted to put in one more opinion on the Cinque Terre question. While I found the area interesting, I am not so entranced that I need to hurry back. This past year, I wanted to take my husband to a very lovely festa in Monterosso al Mare, but we couldn't get a room.
So instead, we spent an extra two nights in my favorite town in Liguria, Santa Margherita Ligure. There are wonderful places to stay, good hikes and boat excursions, and many terrific places to eat.
There are tourists, as the town is a resort, but it is just charming and really a lot of fun. After three visits, I am firm about SML being my favorite town in Italy.
The train connections are very easy from Milan to SML and most of the Ligurian towns. I love this part of Italy and I'm sure you will, too, no matter which town you choose to stay in.
I also second Linda's suggestion of staying in Umbria, near to Assisi and Perugia. They are famous and visited for good reasons.
