Just a couple of weeks ago I took a train from La Spezia to Monterosso ... and as well, a train back through the villages, stopping at Riomaggiore and then back to La Spezia. These local trains run about every 1/2 hour. Most of the trains stop at each of the five villages; however, look at the schedule carefully, as some trains don't stop at all the villages. There's no need to reserve .. and the fares are very modest. FYI - The train stations in both Monterosso and Riomaggiore are well-located (I think) on a level close to the water. There are inclines and steps to get into the 'hearts' of both villages.

Next time I am there I would definitely take a local ferry along some part of the coast ... would recommend that you investigate. (I didn't have time to ferry.)

For your one day CT adventure, I'd suggest taking the train, early in the day, from La Spezia to Monterosso. Work your back to Riomaggiore with a combo of local ferries, hiking and regional trains. Plan your stops around morning coffee, lunch and late afternoon tea/wine. Your fitness level will help you to determine which, if any, of the hiking trails you would like to explore.
