Rome to Florence to Venice? If you don't have a rental car (about 600 km, all the trip on motorways but some spots are very busy) the best way is boarding a Eurostar train. It's the fastest train, with very few stops in between, and it costs just a bit more than the other trains. The ticket includes the seat reservation, so no trouble at peak times, and good space for storing your baggage. When heading to Switzerland from Venice, I suggest a night train with sleepers.
