When we were there in early October, we called the Uffizi reservation number and had no problem making reservations in English. You just need to arrive a bit early to pick up the tickets. The line without reservations was horrendous! If you are going during the summer, I would follow Joel's suggestion that you book well in advance. We were able to get in with just a couple of days notice in October, but I would not take that chance during summer. This is also all true for Galleria Accademia where the David is located.
BTW, one of our group was in a wheelchair, and he was allowed in free with a companion. They were also very accommodating to him.
