I'm going to second P. Miller's advice and vote for Charleston. My wife and I spent a couple of days there two month's ago and really enjoyed it. We had spent the previous week wandering in Virginia, then followed Charleston with a drive to Savannah (another nice walking city), then a drive to Asheville, NC, before circling back to Washington, DC.
We stayed at the King Charles (which I think I reserved through SideStep.com) and found it to be reasonably priced with a great location. We ate at Jestine's Kitchen one night, which was about a block away. Although my wife's fried chicken was great, my meal of dried-out fried pork chops with tasteless macaroni & cheese and okra gumbo side dishes was forgettable. We had a great breakfast of stuffed french-toast one morning, however. (Hell of a diet, eh?)
If you decide to go, pick up a copy of the AAA guide for South Carolina - it has a couple of walking tours of the city that were great.
