Dexter House looks a little out of the way. On 86th close to West End Ave. You're close to one subway line, the 1,2,3. A fair walk to Central Park and the Natural History Museum (but is walkable).
Sandy's suggestion of staying in Times Square is a good one for most people. Lots of transportation there, bus and subway. Close to Broadway shows. Also close to Central Park, 5th Avenue Shopping and other sights like Radio City Music Hall, Rockefeller Center, FAO Schwartz, etc. Hell's Kitchen is starting to get quite a few good restaurants too.
Also, if you get the urge to cab it anywhere, Times Square is much more centralized.
