I think Gavin mentioned Montalcino, and I think that is a wonderful town. When we were there, we ate at the restaurant in the Hotel Il Giglio, which is located right in the town. I did not see the hotel, but the restaurant was good. It is a three star hotel, and you can find information at www.tuscany.net by clicking on Montalcino.
Of course, southern Tuscany is known for all of the vineyards, but Montalcino is close to San' Antimo, and would not be a difficult drive to Montepulciano and Pienza. It is smaller than Montepulciano, and a little bit more accessible.
