First the obvious: Don't forget to get up and walk around regularly - de rigeur on a long flight. Maybe check out a book of office/airline in your seat exercises to add to this routine?
I don't know what airline you're flying on so don't know if you'll have the choice of your entertainment on the video screens in each seat back on such airlines as Virgin Atlantic or if it's a matter of the movie the airline has chosen for you. If you have a Walkman of either tape or CD variety, perhaps a few favorite albums and/or books on tape. Perhaps you can find ones you like at the used book store so you could leave the tapes/CD as a gift and not have to tote them back.
Friends have said Gameboys help pass the tedium, too.
