I definitely would encourage you to rent a car in Tuscany.

You won't be able to drive into many of the Tuscan towns, but rather park your car in a public parking lot outside the city walls and then walk in. This is true of Siena as well. I've not been to the Amalfi coast so, no comment on that part of the trip.
Have you been to Rome before? If not, one day is really not enough. Candice NYC
