I think both the Rijksmuseum and Anne Frank House in one day are doable. Amsterdam is relatively compact. At the Centraal Station tourist office (called VVV) you can get a day pass that I believe includes museum passes and an all day tram ticket. Check out this map: http://www.amsterdam.info/map/

Centraal Station is the red rectangle in B4-B5. The "Museumplein" area (C3) is where you can find the Rijksmuseum, Van Gogh Museum, and the Stedelijkmuseum (modern art). They are literally all next door to each other in a 2-3 block area. You can take Tram 2 out of Centraal Station to get there. I don't remember the exact name of the stop but the tram driver usually calls out the museum names. Also when you get on a tram you can tell the driver where you want to go so s/he is sure to announce it in English (they do this often, don't be shy about asking). I used to live practically next door in C2 on Overtoom off Vondelpark. sigh... those were the days :-)

The Anne Frank House ("Huis" in Dutch pronounced same as in English), is near the Westerkerk (West Church) in B3 off of Prinsengracht, one of the major canals. Take Tram 13 or 17. Link to Anne Frankhuis http://www.annefrank.org/content.asp?pid=1&lid=2
